Ondanks het feit dat de lichtmicroscoop een van de oudste wetenschappelijke instrumenten is, zijn er nog steeds nieuwe ontwikkelingen rond dit instrument. Voor biologische toepassingen speelt met name de fluorescentie-microscopie een belangrijke rol. In fluorescentie-microscopie wordt gebruik gemaakt van moleculen die blauw of UV licht absorberen en vervolgens bij een langere golflengte (roodverschuiving) licht uitzenden. Er kunnen bv fluorescerende moleculen gebruikt worden om selectief cel-organellen te labelen, ionen-concentraties te kwantificeren of membraampotentialen te bepalen. Het gebruik van fluorescerende ‘probes’ is echter niet geheel zonder problemen.
